I think your pumpkin plants look good. The tissue tests may or may not help identify the issue it could just be old age and normal transplant stress. If the growing tips look good, you don't have to worry too much about the older leaves. If you're on a budget, I'd say save your money for when the growing tips start to look funky. I think the tissue tests might be a better tool for solving issues with new growth than issues with the old growth. But that's just one opinion and I'm sure other growers would have a different opinion.
Tomatoes look good... Good luck with them.
