I heard that it does not since an Jack O' Lantern is a C. Pepo and a Ag is a C. Maxima

You could use strawberry pollen and it'd take. It doesn't take a specific pollen to pollinae a pumpkin, unlike other plants.
In 2008 I used AG pollen to pollinate my 'Unknown Pumpkin' (Which turned out to be a Jack o Lantern)
Obviously if you use Jack o Lantern pollen to pollinate a AG, the offspring would most likely only be around 50Lbs, but if you don't care about offspring then...Yes!

If your talking about the variety Jack O Lantern,it is c.pepo. C.pepo and C.maxima can't hybridize.

Oh wait a minute,, I used belly button lint one time to pollinate my AG and it worked just fine,,,,,my bad,, forgot about that.

I thought all the cucurbits would cross pollinate. Does anyone have a link to an article that discusses the limits and extents of cucurbit x-pollination? Oh, such an educated group! Mr. d&me - how about a cross with a pot plant - than we could all retire! Except it would be nuts trying to keep the hippies out of your patch. Thanks M

Ill tell ya who would be really a really good expert in explaining all this and thats ether Joe Ailts or Ron Rahe,they will set you guys straight when it comes to this category.
But a C.Pepo (Field Pumpkin) and a C. Maxima (AG) will not cross.

Must be the summer heat has fried some people brains. In this world today, no one has a sense of humor. Can't brake anyone bubble with out anyone having a fit or flying off the handle. Now Carrie, If a pumpkin is open pollinated by bees ( from a apple, pear, flowers)and it works, then now why wouldn't a Jack O lantern pumpkin work, Even a blonde should be able to figure that one out. Went to a cookout yesterday and some joker flew off the handle at a joke and started a fight. Poor guy won't ever be invited to a 4th July cookout again. Everyone was right there and was laughing at the joke. Some people just need to have a life and take a good joke once in a while.Now you see why I never hardly post or reply to post anymore.

Cary You can't cross different geno types with pollen, As some of the others have answered, Giant pumpkins are Maxima, and will only cross with other maxima pumpkins or squash. The Pepo geno type will on very specific occasions take pollen from a Maxima, somewhere in the 1,000,000 to 1 but not the reverse, and that is because of the DNA strains.

Squash and Pumpkins
Cucurbita maxima
C. pepo
C. moschata
C. mixta
Squash, like beans, corn and tomatoes were domesticated in Central America. Seeds have been discovered in the Andes and in Mexico that carbon dated to 8,000 B.C., making squash one of the oldest domesticated crops. Squash spread north into North America and became an important crop for the native peoples living there. To clear up the biology involved, pumpkins are simply round orange squash, regardless of the species. Squash and Pumpkins cross-pollinate readily with other members of their species, but not with different species. Therefore, a spaghetti squash (C. pepo) will cross with an acorn squash or even a zucchini, but not with blue hubbard (C. maxima) or nippon island (C. mixta). :souce Annapolis seeds

Brooks, thanks for the nod. Cary, Ed's post is spot on. It is biologically impossible to cross a jack-o-lantern with an AG pumpkin.
This is the equivalent to chimpanzees and gorrillas trying to reproduce...they look alike but are just different enough that it cannot work.
